Popis: | Cytoplasmic starch phosphorylase from yam ( Dioscorea cayenensis ) was partially purified by ammonium sulfate fractionation. chromatography on DEAE A-50, Sephadex G-100 and Sepharose 6B filtration. Only one band showing phosphorylasic activity was found upon polyacrylamide gel electrophoresis. Its apparent molecular weight was 200 000 ± 3000. The enzyme is specific for starch and glucose 1-phosphate; it requires starch for its activity in the synthesis way. The enzymatic activity was inhibited by Hg 2+ and EDTA at high concentrations. Glucose did not inhibit the reaction. Thiol and Tyrosyl groups are required for enzymatic activity. |
