Popis: | To the Editor:—Sachs et a/.'s (96:381384, 1991) paper about false-positive reactions in cryptococcal antigen determination induces me to make some remarks. The problem of false-positive reactions in the test-based on latex agglutination is well known, especially when the antigen to be detected does not allow pretreatment with heat enzymes that deactivate the rheumafactor (perhaps the main offender in false-positive reactions, but not the only one). Dr. Zaini and co-workers at the University of Teheran have found in their studies of false-positive reactions in cryptococcal antigen tests that the false-positive reaction correlates with the iron content in the tested sera. The mechanism seems to be Fe reacting with the azide ion and/or negative charge of the antigens bound to the latex particles. Sera above 200 /ug Fe/dL gave false-positive reactions. 2-Merkaptoethanol reduces Fe to Fe and eliminates the phenomenon. There are still new pieces to be added to the puzzle of false-positive latex agglutination reaction. |
