Popis: | SDS-PAGE and RP-HPLC imply a total reduction of native protein aggregates. This leads to a loss of information concerning structure, interaction and stability of protein complexes. A major advantage of SE-HPLC is its potential to work with relatively large aggregates, and it is more likely to approach the physicochemical and structural basis of wheat quality. The predictive value of SE-HPLC for breadmaking quality in a single locality and across localities was investigated. SDS was used for the first protein extraction step, followed by sonication to extract the remaining protein. SDS soluble gliadins were positively correlated to quality, irrespective of the locality. In a single environment, the insoluble polymeric proteins had no significant effect on quality, but when quality was measured in different environments; the SDS insoluble large polymeric proteins had a significant positive effect on quality.
