Biotin-deficient rat liver mitochondria showed decreased Ca 2+ : O ratio as compared to normal rat liver mitochondria when NAD + -linked substrates were oxidized, but not when succinate was oxidized, indicating a damage at the energy conservation site I. 2. 2. Succinate-linked reduction of acetoacetate using aerobically-generated high energy intermediate was also decreased in the biotin-deficient rat liver mitochondria. 3. 3. Administration of 200 μg. of biotin to deficient animals restored the succinate-linked reduction of acetoacetate.
