1 The activation of brain adenylate cyclase by Ca++ in association with a Ca++-dependent regulator protein (CDR) is described. Adenylate cyclase and CDR were solubilized with Lubrol-PX and then separated from each other with the use of DEAE-cellulose chromatography. 2 Solubilized adenylate cyclase was activated rapidly by Ca++ in the presence of CDR. This activation was readily reversed in the presence of an excess of EGTA. 3 Membrane-bound adenylate cyclase from rat brain exhibited striking regional differences in sensitivity to Ca++. These differences disappeared upon solubilization of the preparations. 4 A model is presented which proposes that cellular cyclic nucleotide levels are regulated by free Ca++ via a specific Ca++ receptor. |
