Popis: This chapter discusses the basic components of a high performance liquid chromatography (HPLC) system. Attention is given to origins of chromatographic analysis and recent advances in the field along with related HPLC techniques. The primary focus of the chapter regards the instrumental setup. Components of the instrument discussed in detail include mobile phase/solvent reservoir, solvent delivery system, sample introduction device, column, postcolumn apparatus, detectors, data collection and output, postdetection eluent processing, and connective tubing and fittings. An overview of various HPLC detectors is provided with discussion of unique detector characteristics and a comparison of advantages and drawbacks between them. Focus is placed on the most common detectors, including UV/Vis absorbance, fluorescence, electrochemical, conductivity, refractive index, and mass spectrometry detectors. This chapter provides a basic working knowledge of all instrumental components required for HPLC analyses.
