Isolation of adult bone marrow and fetal liver cells from murine models is an important technique that can be a valuable tool to study mechanisms of hematotoxicity. Organic solvents are a class of industrial chemicals that can target the bone marrow leading to hematatoxicity including leukemia. Many of these chemicals are bioactivated into reactive intermediates, which can initiate oxidative stress; thus, identification and quantification of oxidative stress is essential to gain further understanding of the mechanisms of organic solvent-initiated toxicity. This chapter describes the procedure for isolating bone marrow from adult mice and for isolating fetal liver cells, which are the primary site of hematopoiesis in the developing embryo. Additionally, descriptions of molecular techniques that can be employed to identify and quantify oxidative stress caused by exposure to organic solvents are included. |
