GLAD-PCR assay of methylation of ACGC site in position Chr11: 65647266 in DNA samples from the blood cells of healthy donors and the early-stage renal cancers

Popis: Methylation of ACGC site in position Chr11: 65647266 (according to GRCh38 PrimaryAssembly) in DNA samples from blood cells of healthy donors and early stage renal cancer patients was studied with GLAD-PCR assay. The work includes a) preparation of light fraction of the blood cells, b) isolation of DNA from a light fraction of the blood cells and determination of DNA concentration by real-time PCR, c) determination of A(5mC)GC site (Chr11: 65647266) concentration in DNA preparations by GLAD-PCR assay and d) calculation of percent of DNA molecules with A(5mC)GC site in position Chr11: 65647266. It was shown that a methylation of ACGC site (Chr11: 65647266) in DNA preparations from a light fraction of the blood cells of the early stage renal cancer patients is varied from 96 to 126 . At the same time this site methylation in DNA preparations from a light fraction of the blood cells of conditionally healthy donors lays in a range 72 85 . Thus, GLAD-PCR assay of A(5mC)GC site (Chr11: 65647266) in DNA preparations from a light fraction of the blood cells allows to exclude early stage renal cancer. GLAD PCR assay doesnt require special and expensive equipment. The study can be carried out in a standard PCR laboratory when a blood analysis is taken place.
