Combined multiple genes phylogenetic analysis and morphological characteristic description of entomopathogenic fungi infecting cicada nymph from northeast of Thailand

Popis: | Recently, entomopathogenic fungi that infect cicada nymphs (Megapomponia intermedia and Pompania platylomia) have been reported to be used in traditional medicine, such as to treat asthma, bronchial and lung inflammation and kidney disease. In Thailand, these entomopathogenic fungi have been found in various locations; however, species identification has not been well studied. Therefore, the aim of this study was to investigate the fungal species from 44 strains of cicada nymphs’ entomopathogenic fungi using a combined multiple genes phylogenetic analysis. The results indicated that this combined data set of internal transcribed spacers of nuclear ribosomal DNA repeats (ITS), small subunit of (nrSSU) rDNA and large subunit of (nrLSU) rDNA, and two coding genes consisting of the elongation factor 1α (EF-1α) gene and the largest subunit of the RNA polymerase II (rpb1) gene had clearly identified 31, 4, 5, 3 and 1 isolates as coming from Polycephalomyces nipponicus, Ophiocordyceps longissima, Simplicillium obclavatum, Metacordyceps chlamydosporia and O. sobolifera with 0.9–2.8%, 1.4–3.7%, 0.1%, 0.2% and 3–8% genetic distances, respectively. Finally, the morphological structures of these fungal species, such as stroma, perithecia, asci, ascospores and part-spores, were described in this study. |
