Popis: | A simple method for measuring the changes of the optical density with time, after heating or cooling a solution within seconds, is described. The temperature-changes in the solution are achieved by switching the circulating fluids of two thermostats alternating through a microcell. Possible applications of this method are discussed. The kinetics of the reversible denaturation of α-Chymotrypsin in the pH-range of 2.0–2.5 were measured. These measurements are in agreement with a transition between two different states of the protein molecule and allow the evaluation of the rate constants. The temperature dependence of these rate-constants is presented and discussed by a qualitative picture of the denaturation process. |
