Popis: Prompted by the observation that the lack of CD95 surface staining in pancreatic tumor tissue as well as the low surface expression on pancreatic carcinoma cell lines correlated with both high resistance to CD95-induced apoptosis and strong expression of PKCμ, we analyzed the possible role of PKCμ as an anti-apoptotic factor in CD-95-induced apoptosis. Inhibition of PKCµ by Go6983 dramatically enhanced apoptosis in PancTuI, Panc89 and Colo357 cells in response to anti-CD95 Stimulation. Stable overexpression of PKCμ in Capan 1 cells, though not altering total CD95 protein levels, strongly decreased CD95 cell surface staining and, in parallel, sensitivity to CD95-induced apoptosis. These data suggest that PKCμ may exert its anti-apoptotic function, at least in part, by reducing the fraction of CD95 on the cell surface.
