Knowledge about nitrogen (N) cycling in aquatic ecosystems is limited. While the dinitrogen fixation (N2-fix) process is comparably well studied, the contrasting denitrification (DENI) pathway is highly under-investigated, because of expensive and relatively complex experimental techniques currently available. Thus, we present a combined acetylene assay termed COBRA (= combined blockage/reduction acetylene assay) for the simultaneous determination of N2-fix and DENI in aquatic samples. The technique measures gaseous headspace samples via gas chromatography. Both target gases that accumulate (ethylene for N2-fix, nitrous oxide for DENI quantification) during incubations can be detected and quantified in a single assay and from a single sample. |
