Use of Tn phoA to tag exported proteins in Myxococcus xanthus

Popis: Tn phoA was introduced into Myxococcus xanthus, and insertion mutants that expressed the E. coli alkaline phosphatase (PhoA) were selected. These mutants displayed hybrid proteins of various sizes containing PhoA antigenicity. The transposed strains expressed different levels of E. coli alkaline phosphatase activity; in addition they varied in the fraction of extracellular enzyme from mutants with mostly cell bound activity to mutants that produce the phosphatase in the extracellular medium. Therefore sequences coding for signal peptides should exist in M. xanthus, and Tn phoA can be a suitable tool to study the events involved in protein secretion in M. xanthus.
