One of the most popular routes of drug administration is the oral route because of its ease of administration. However, it does not fit well for all therapeutic agents. Therefore the buccal region of the oral cavity is an attractive target for drug administration and can overcome drawbacks associated with oral drug delivery systems like high first-pass metabolism and acid instability of the drug. Moreover, rapid onset of action can be achieved and discontinuation of therapy is easier. Buccal drug delivery is a promising drug delivery system for drugs that cannot be administered by oral means, and it is also an attractive noninvasive delivery means for peptides and other proteins. This chapter focuses on oral mucosa anatomy, absorption pathway, barriers to penetration, advantages and disadvantages of buccal drug delivery systems, means to circumvent the disadvantages, potential drug candidates and formulation design of a buccal delivery system, natural and synthetic mucoadhesive polymers, marketed preparations, current development, and future aspects. |
