Development of microsatellite markers from four Pittosporum taxa (Pittosporaceae) endemic to the Ogasawara (Bonin) Islands with cross-amplification in Ogasawara Pittosporum and P. tobira

Popis: A total of 55 microsatellite loci were developed and characterized using four Pittosporum taxa endemic to the Ogasawara (Bonin) Islands, Japan: Pittosporum boninense var. boninense, P. boninense var. chichijimense, Pittosporum parvifolium var. parvifolium and P. parvifolium var. beecheyi. Transferability of these markers among the Ogasawara Pittosporum taxa as well as Pittosporum tobira was also evaluated. The number of alleles per locus ranged from 2 to 23 in P. boninense var. boninense, from 2 to 16 in P. boninense var. chichijimense, from 1 to 7 with in P. parvifolium var. parvifolium, and from 1 to 8 in P. parvifolium var. beecheyi. During cross-amplification tests, the majority of these loci were successfully amplified and showed moderate to high levels of genetic variation. The markers described here could be used to achieve a more accurate determination of genetic diversity and genetic structure of the taxa from which they were derived.
