Popis: The study of the interaction of antipsychotic drugs with central neurotransmission has implicated serotonin at least as early as dopamine. The interest in serotonin antagonism was not sustained because central dopamine D2 antagonism was found to be the common activity of all antipsychotics and clinical studies with methysergide and cinanserin were disappointing. The development of selective 5HT2 antagonists devoid of partial agonist activity revived the interest in the physiological and pathological role of serotonin. Ritanserin and risperidone have applications in psychiatry by virtue of potent, neurologically safe 5HT2 antagonism.
