A Hypoxanthine Biosensor for Estimation of The Postmortem Interval

Popis: This chapter presents an overview of a hypoxanthine (Hx) biosensor for estimation of the postmortem interval. An Hx biosensor consisting of a xanthine oxidase immobilized membrane and a hydrogen perioxide electrode was developed and applied for estimation of the PMI. The enzyme was covalently immobilized on a membrane prepared from cellulose triacetate. A model SBA-40 analyzer was used, consisting of a 500 μl chamber equipped with hydrogen peroxide electrodes. The electrode responded linearly to Hx in a range 6.25–250 μM at pH 7.2. The coefficient of variation was 0.59% for 20 repeated assays. More than 1000 assays could be performed with the same membrane and each sample could be assayed in 20 s. The membrane could be stored at 4°C for 12 months. When applied to the determination of Hx in the vitreous humor of rabbit, the results obtained agreed well with those obtained by the conventional enzymatic method. The results suggested that Hx biosensor provides a new method for the estimation of the PMI.
