Systemic tumour necrosis factor (TNF) treatment in psoriasis: clinical data from a total of 14 patients in an open trial

Popis: We have previously reported a beneficial effect of systemically administered recombinant human tumour necrosis factor (TNF) in two out of three patients with intractable psoriasis. To supplement the preliminary report, we have compiled data from all of the 14 psoriatic patients treated so far in an open trial of systemic TNF. Nine of the 14 patients were treated with doses of 2 × 105 U/m2 body surface for 5 days, 3 × 105 U/m1 for 5 days and 5 × 105 U/m2 for 5 days with 2 weeks with no TNF treatment between each successive dose increase. The other five patients were treated with 1–2 × 105 U/m2 TNF 6 days per week for 8 weeks. Because systemic side-effects including fever, chill or shivering during the infusion of the drug were unavoidable, two patients dropped out of the study.
