Popis: | The electron microscopic extraction of information from biological specimens has always been a compromise among at least four factors: resolution, contrast, radiation damage and signal-to-noise ratio. To overcome the notoriously low contrast of most biological specimens examined in bright field, heavy atom staining or shadowing has been used as a seemingly inevitable necessity. Three benefits result, the most important being that one can now observe structure. In addition, however, the specimen, or rather its heavy atom shell, is relatively radiation resistant and can be examined at length. Nevertheless,Williams and Fisher (1970) have indicated the importance of low specimen exposure even under these conditions. |
