Popis: | Nuclear morphology studies are a reliable parameter for analysis of genotoxic properties of different agents. Such studies have been widely exploited in isolated protoplasts but seldom used in cells isolated from whole root tissue due to cell wall permeability restrictions. We present a rapid and easy protocol that will benefit the researchers in studying the nucleus stained with fluorescent dye. Here, we have modified the digestion buffer and used a permeabilization buffer to ease the entry of the fluorescent Hoechst stain through the cell wall, so that fluorescence can be observed under UV light. |
