Fundamental objective of proteomic study is to identify and quantify maximal number of proteins in any given sample. Identification itself is much easier to accomplish using state-of-the-art mass spectrometers with high sensitivity and resolution capabilities. Quantification is much more demanding task and needs to satisfy two criteria: accuracy and precision. Therefore several analytical approaches have been developed for quantitative proteomics. Some of them are based on labeling, metabolic or chemical, some of them are label free techniques. All techniques that are in current use have been described here including gel-free-based quantitative proteomics, metabolic labeling, chemical and label-free techniques. |
