Effect of Melatonin and Salicylic Acid on ROS Generation by Mitochondria of Lupine Seedlings

Popis: The effect of melatonin and salicylic acid (SA) on respiration and ROS (hydrogen peroxide) generation during the oxidation of succinate by mitochondria isolated from cotyledons of 5-day-old etiolated lupine seedlings (Lupinus angustifolius L.) was investigated. It was shown that melatonin did not have a noticeable effect in a wide range of concentrations (10–7–10–4 M) on the rate of succinate oxidation in various metabolic states and the amount of respiratory control, but, depending on the concentration, it significantly (up to 60% of control) inhibited the formation of ROS by mitochondria, the main contribution to which was made by complex II (succinate dehydrogenase). The presence of 0.05–0.5 mM SA in the incubation medium activated (by 1.5–2.5 times) the formation of hydrogen peroxide during the oxidation of succinate. At the same time, SA at low concentrations had a predominantly uncoupling (activating) effect on the oxidation of succinate in state 4 (in the absence of ADP), and respiration was inhibited in state 3 (in presence of ADP) with an increase in its concentration. In general, the results obtained demonstrate the ability of melatonin and SA to exert a direct regulatory effect on the generation of ROS by mitochondria, protecting organelles from oxidative stress, as well as promoting their participation in the formation of plant response to unfavorable environmental conditions.
