PRELIMINARY RESULTS OF SOMATIC EMBRYOGENESIS FROM YOUNG ZYGOTIC EMBRYOS OF OLIVE TREE

Popis: | In Tunisia, as in many Mediterranean countries, olive growing plays a strategic economic role at it makes possible the exploitation of the poorest soils of the southern regions. In the present work, two major cultivars, Chemlali of Sfax and Meski, were used. Callogenesis was induced easily from many types of tissues, i.e. explants taken from young or adult trees, but none showed induction of somatic embryos. Besides, young zygotic embryos were collected 20, 30,40, 120 days after full bloom and tested for their somatic embryogenic capacity. The embryos were grown on MS medium and kept in the dark for 6 weeks followed by 16 h photoperiod for two months. Somatic embryogenesis was observed on 70-day-old zygotic embryos in medium containing either low cytokinin concentration or the combination NAA and 2iP, with a rate reaching in some cases 30%. The use of active charcoal seems to improve the embryogenic capacity of immature zygotic embryos. |
