Popis: The interactome is defined as the entirety of interactions between biological macromolecules of a cell comprising the full spectrum from purely functional relationships to direct physical interactions between them. It is this complex web of interactions that drives all biological processes and can be considered the scaffold of cellular systems. We will overview different strategies of biophysical protein-centric interactome mapping and discuss exciting findings obtained from integrating biophysical with functional interaction maps that spurred understanding of cellular organization and its perturbation upon disease.
