Effect of GLP-1 agonist on osteoblast proliferation and related signaling pathways

Popis: Objective: To study the effect of glucagon-like peptide-1 (GLP-1) agonist on osteoblast proliferation and related signaling pathways. Methods: Osteoblast cell lines MG-63 were cultured and divided into three groups, GLP-1 group were treated with the medium containing GLP-1, EX-4 group were treated with the medium containing GLP-1 analogue, and control group were treated with serum-free medium. The expression levels of proliferation genes BMP-2, Dlx2, PCNA and Caspase-3, Wnt pathway molecules Wnt1, Wnt3a, Wnt5a, GSK- 3β, β-catenin and Runx2 as well as Hedgehog pathway molecules Shh, Ihh, Smo, RANKL and OPG were measured after 24 hours. Results: BMP-2, PCNA, Wnt1, Wnt3a, Wnt5a, β-catenin, Runx2, Shh, Ihh, Smo and OPG mRNA expression in GLP-1 group and EX-4 group were significantly higher than those in control group whereas Dlx2, Caspase-3, GSK- 3β and RANKL mRNA expression were significantly lower than those in control group, and the expression levels of proliferation genes, Wnt pathway molecules and Hedgehog pathway molecules in GLP-1 group were not significantly different from those in EX-4 group. Conclusion: GLP-1 agonist can promote the osteoblast proliferation and enhance the activity of Wnt pathway and Hedgehog pathway.
