Bakterije roda 'Candidatus Phytoplasma' su unutarstanični, biljni patogeni bez stanične stijenke iz razreda Mollicutes. Uzrokuju bolesti brojnih biljaka širom svijeta. Svrha ovog rada bila je istražiti učinak egzogenog biljnog regulatora rasta indol-3-maslačne kiseline (IBA) na bakteriju 'Candidatus Phytoplasma pruni' (soj KVI) i domaćinsku biljku madagaskarski zimzelen (Catharanthus roseus (L.) G. Don). Zdravi i bakterijom zaraženi izdanci madagaskarskog zimzelena dugoročno su održavani u kulturi tkiva in vitro na hranidbenoj podlozi s dodanim biljnim regulatorom rasta indol-3-maslačnom kiselinom (IBA) ili benzilamunopurinom (BAP). Tijekom tri supkulture (13., 14. i 15.) mjerene su i uspoređivane visine izdanaka. Rezultati su pokazali da je biljni regulator rasta IBA povoljno utjecao na produžni rast fitoplazmom zaraženih izdanaka madagaskarskog zimzelena. Prisutnost bakterije 'Candidatus Phytoplasma pruni' (soj KVI) u zaraženim izdancima madagaskarskog zimzelena provjerena je „ugniježđenom“ lančanom reakcijom polimerazom. Fitoplazma nije detektirana u 83% izdanaka tretiranih IBA-om, što pokazuje da IBA ima učinak na tu bakteriju. Bacteria of the genus 'Candidatus Phytoplasma' are intercellular, wall-less plant pathogens from the class Mollicutes. They cause diseases in numerous plants worldwide. The purpose of this study was to investigate the effect of the exogenous plant growth regulator indole-3-butyric acid (IBA) on the 'Candidatus Phytoplasma pruni' (strain KVI) and the host plant Madagascar periwinkle (Catharanthus roseus (L.) G. Don). Healthy and infected shoots of Madagadscar periwinkle were grown in vitro on the nutrient medium containing growth regulator indol-3-butyric acid (IBA) or benzylaminopurine (BAP). During three subcultures (13., 14. and 15.) heights of their shoots were monitored and compared. The results have shown that plant growth regulator IBA had positive effect on the elongation of phytoplasma-infected Madagascar periwinkle shoots. The presence of 'Candidatus Phytoplasma pruni' (strain KVI) in the infected shoots of the Madagascar Periwinkle was tested by nested Polymerase Chain Reaction. Phytoplasma was not detected in 83% of the shoots treated with IBA, which shows that IBA has the effect on that bacterium. |
