Popis: | Zbog nepovoljnog igličastog oblika kristala memantin hidrogen klorida koji je farmaceutski aktivna tvar koja pokazuje aktivnost u liječenju Alzheimerove bolesti, bilo je potrebno naći sol povoljnijeg oblika kristala. Na temelju istraživanja djelovanja i usporedbe s memantin hidrogen kloridom, zbog znatno bolje morfologije kristala i higroskopnosti izabran je memantin hidrogen sulfat. U okviru ovog rada, u procesu šaržne kristalizacije, istražen je utjecaj procesnih parametara te dodatak pomoćnih komponenti (antiotapalo i cjepivo) na granulometrijska svojstva memantin hidrogen sulfata. Vrsta otapala i način vođenja procesa utječu na prezasićenost sustava, termodinamiku i kinetiku kristalizacije te granulometrijska svojstva kristala. U svrhu proizvodnje produkta odgovarajućih granulometrijskih svojstava, u ovom istraživanju provedena je šaržna kristalizacija memantin hidrogen sulfata hlađenjem iz 2-propanola uz dodatak antiotapala (etilacetat i n-heptan) i dodatak cjepiva unutar metastabilne zone. Nakon završetka procesa kristalizacije napravljen je filtracijski test, a kinetika sušenja određena je iz podataka dobivenih sušenjem produkta u atmosferskom sušioniku. Oblik kristala praćen je mikroskopskom analizom, a metodom laserske difrakcije određena je raspodjela veličina kristala. Kinetika procesa opisana je pomoću odgovarajućih potencijskih jednadžbi. Memantine hydrochloride is pharmaceutically active substance that was found to have role in treatment of Alzheimer's disease. Because needle-shaped crystals of mementine hydrochloride were not appropriate for further use in this particular case it was necessary to find another salt of more suitable formation. After reviewing previous research, memantine hydrogen sulphate was chosen since it has significantly better crystal morphology and hygroscopic behaviour. In this research, through the batch crystallization process, the influence of process parameters and addition of auxiliary components (antisolvent and seed) on granulometric properties of memantine hydrogen sulphate was investigated. Type of solvent and process management influence the saturation of the system, thermodynamics, crystallization kinetics and granulometric properties of crystals. Since the aim of this study was to create a product of appropriate granulometric features, batch crystallization of memantine hydrogen sulphate by cooling from 2- propranol with addition of antisolvent (ethyl acethate and n- Heptane) and seed within metastable zone was performed. After terminating the crystzallization process, filtration test was performed and drying kinetics was determined from information derived from products drying in atmospheric dryer. The shape of crystals was monitored with microscopy analysis and the distribution of crystal size was determinated using laser disfraction method. Process kinetics was described using appropriate potential equations. |
