Balıklarda list deriden salgllanan mukus SlVIS1 ad1 verilen kaygan madde, deriden viicudagirebilecek mikroorganizmalarm giri~ini engelleyen bir tabakadrr. Bu yah~ma, ballklarmkontamine ortamlarda saglIkll kalabilmelerini saglayan faktorler arasmda mukus SIVISllllnroliinli belirlemek amaclyla yap1lmt~trr. Sivas, Ul~ Karacalar Baraj Golli ve Tokat, AlmusBaraj Golli'nde bulunan Salmo, Cyprinus ve Leuciscus cinslerine ait ballklarm mukusslvdan almm1~ ve 3 farkh solvent (Dirnetil siilfoksit, Etanol ve Kloroform) kullamlarakekstrakte edilmi~tir. Elde edilen mukus ekstraktlannm antibakteriyel ve antifungalozellikleri ar~tmlrru~trr. Omekler, 11 bakteri ve 8 fungus olmak iizere 19 farkllmikroorganizma iizerinde denenmi~tir. Bu 9ah~mada kullamlrru~ olan gram pozitifbakteriler (Staphylococcus aureus, Bacillus subtilis, Bacillus cereus), gram negatifbakteriler (Eschericia coli, Pseudomonas aureginosa, Klebsiella pnemoniae, Enterobacteraerogenes, Salmonella enteritidis, Salmonella gallinarum, Enterococcus lecalis, Proteusvulgaris) ve funguslar (Aspergillus flavus, Aspergillus lumigatus, T roseum, Penicilliumbrevi-com pactum, Alternaria alternata, Candida utilis, Penicillium purpurogenum,Aspergillus niger) iizerindeki biyolojik aktiviteleri Disk Difiizyon Metodu, MIC ve MBCmetodlanna gore ~tmlrru~rr. Elde edilen bulgulara dayanarak mukus ekstraktlan iyindeantibiyotiklerin olu~turdugu inhibisyon zonlarma en yakm ve kayda deger etkinin Proteusvulgaris iizerinde oldugunu soylemek miimkiindiir. (Gentamisin 24 mm, SCF 21 mm,Azitromisin 13 mm, Neomisin 15 mm). Yapdan degerlendirmede 2 farkh bolge ve 3 farkllballk cinsinden elde edilen mukus ekstraktlannm pozitif ve negatif kontroller goz onlindebulunduruldugunda biryok bakteri ve fungus iizerinde kayda deger bir biyirnde zonol~turamadtgl goriilm~tiir. Bu nedenle mukus ekstraktlannm herhangi bir biyolojikaktivite gostermedigi sonucuna vanlrru~t1r.Anahtar Kelimeler: Bahk, Mukus, Antimikrobiyal aktivite, Tath su The upper skin of the fish called mucosal fluid, a slippery substance, is known toprevent the entry of microorganism into the skin layer. This study focuses the role ofmucus fluid to protect fish in contaminated environment. The mucus and fluids from thetypes of Salmo, Cyprinus and Leuciscus fish in Sivas, Ulas Karacalar Lake Dam, DamLake Almus taken and extracted by using 3 different solvent (dimethyl sulfoxide,ethanol and chloroform). In mucus, antibacterial and antifungal properties of theobtained extracts were investigated. Samples are tested on 11 to 19 differentmicroorganisms, including bacteria and fungi. In this study gram-positive bacteria(Staphylococcus aureus, Bacillus subtilis, Bacillus cereus), gram-negative bacteria(Escherrischia coli, Pseudomonas aureginosa, Klebsiella Pneumonia, Enterobacteraerogenes, Salmonella enteritidis, Salmonella gallinarum, Enterococcus Ie calis,Proteus vulgaris) and funguses (Aspergillus jlavus, Aspergillus lumigatus, T roseum,Penicillium brevi-com pactum, Alternaria alternata, Candida utilis, Penicilliumpurpurogenum, Aspergillus niger) are tested in terms of their biological activity throughDisk Diffusion Method, MIC and MBC. Based on obtained findings, when we comparewith inhibition zones formed by antibiotic, it is possible to say that the closest andremarkable effect is on Proteus vulgaris in mucus extracts (Gentamicin 24 mm, SCF 21mm, Azithromycin 13 mm, Neomycin 15 mm). The findings show bacteria and fungiwere not able to generate a significant way zone when mucus from fish extracts in twodifferent regions and three different types are considered. Therefore it is concluded thatmucus extracts do not show any biological activity.Key words: Fish, Mucus, Antimicrobial activity, Fresh water 80 |
