Popis: | Cilj ovog rada je istražiti sposobnosti različitih ekstrakata i čistih spojeva koji dolaze kao sastavnice ekstrakata u inhibiciji dvaju enzima važnih u patogenezi Alzheimerove bolesti; acetilkolinesteraze (AChE) i butirilkolinesteraze (BChE). Predmet istraživanja su bili fenolni spojevi, prvenstveno flavonoidi. S ovim ciljem, pretražene su znanstvene baze podataka (ScienceDirect, Web of Science, PubMed, Google Scholar, PMC i PubChem). AChE i BChE imaju važnu ulogu u hidroliziranju acetilkolina što omogućava prijenos živčanih impulsa. Inhibitori AChE i BChE koriste se kao lijekovi, ali zbog pratećih nuspojava znanstvenici su u potrazi za novim inhibitorima, izoliranim iz biljaka. Rezultati su pokazali da 6-OH-luteolin-7-O-β-D-glukozid, pomiferin, (2S)-5,2-dihidroksi-7,5-dimetoksiflavanon, hispidon, 3-metoksikvercetin i tilirozid imaju značajnu inhibicijsku aktivnost na AChE. Nadalje, od biljnih ekstrakata najbolji rezultat u inhibiciji AChE postigli su etanolni i metanolni ekstrakti korijena biljke Moringa oleifera i frakcije iz korijena biljke Carpolobia lutea (etil acetatna, vodena i metanolna). The aim of this paper is to investigate the ability of various herbal extracts and pure compounds which come as constituents of these extracts in the inhibition of two enzymes important in the pathogenesis of Alzheimer's disease; acetylcholinesterase (AChE) and butyrylcholinesterase (BChE). The subject of the study were phenolic compounds, primarily flavonoids. To this end, scientific databases (ScienceDirect, Web of Science, PubMed, Google Scholar, PMC and PubChem). AChE and BChE play an important role in the hydrolysis of acetylcholine resulting in nerve impulse transmission. AChE and BChE inhibitors are used as medicines, but because of the side effects, scientists are looking for new inhibitors, isolated from plants. The results showed that 6-OH-luteolin-7-O-β-D-glucoside, pomiferin, (2S)-5,2-dihydroxy-7,5-dimethoxyflavanone, hispidone, 3-methoxyquercetin and tiliroside have significant inhibitory activity on AChE. Furthermore, of all plant extracts, Moringa oleifera root extracts (ethanol and methanol) and Carpolobia lutea root extracts (ethyl acetate, aqueous and methanol) achieved the best results in AChE inhibition. |
