The lipid antioxidant vitamin E is among the supplements essential for humans and animals. Its biosynthesis pathways in plants and cyanobacteria have been elucidated and studied, cloned and expressed in order to enhance the vitamin E contents of producer crops. For decades, Escherichia coli has been the primary workhorse for recombinant pharmaceuticals, especially for small molecules and proteins not requiring complex post-translational modifications. This work reviews the biosynthesis of tocochromanols, their functions, and past efforts in engineering the pathway, and the possibilities for E. coli as the optimal host system.
