Evaluación de los posibles efectos sobre la agregación β amiloide y la viabilidad celular tras la exposición con campos electromagnéticos de baja frecuencia

Popis: In recent years there has been a growing awareness of the possible physiological effects caused by the absorption of non-ionizing electromagnetic radiation. This fact is supported by the wide variety of studies that have used electromagnetic energy, particularly extremely low-frequency electromagnetic fields (ELF-MF), as a new therapy, studying its interaction with molecular and cellular parameters in different cell lines. However, the available research on the effects they produce on cells and their possible association with neurodegenerative disorders is contradictory. Therefore, the present study has focused on analyzing the possible effect of this type of fields on the proliferation in vitro of two cell lines, a tumoral, neuroblastoma (N2A), and a non-tumoral, fibroblasts (3T3); in order to design a specific exposure protocol that could be effective in reversing the neurotoxic damage produced by the Aβ1-42 peptide, associated with Alzheimer's disease. The study was performed with an ELF-MF of 1 mT and 75 Hz frequency, using a Helmholtz coil system. Its effect on cell proliferation and viability, on the aggregation pattern of the β-amyloid peptide, and its possible effect in combination with the peptide on cell viability and on the expression of stress proteins were analyzed. Our results indicate that an electromagnetic field of these characteristics, may produce changes in molecular pathways that leads to a reduction of cell growth, more significantly in the tumoral cell line. However, the electromagnetic field did not seem to revert the neurotoxicity caused by β-amyloid peptide.
