Popis: | Karcinom dojke među najčešćim je karcinomima u populaciji žena diljem svijeta od kojeg obolijeva četvrtina žena novooboljelih od raka, te jedan od najvažnijih uzroka smrti. Prema objavljenim studijama incidencija karcinoma dojke varira prema rasi i etničkom porijeklu te je češći u razvijenim zemljama. Cilj ovog istraživanja jest analizirati ekspresiju kalpaina i AR u karcinomu dojke, utvrditi prognostički značaj ekspresije tumorskih markera, razmotriti postoji li mogućnost korištenja AR i kalpaina kao potencijalnih meta odgovora na terapiju. U studiji je sudjelovalo 219 novodijagnosticiranih pacijentica u periodu od siječnja 2000. do prosinca 2004. godine. Kliničko patološki podatci prikupljeni su pomoću baze podataka Zavoda za Patologiju. Medijan dobi prilikom dijagnoze bio je 65 godina. Uzorci su obrađeni tehnikom genske postrojbe (engl.microarray). Blokovi tkiva uklopljeni u parafin koji su sadržavali invazivnu komponentu karcinoma rezani su na rezove debljine 4 μm te su bojani HE tehnikom po standardnom protokolu u svrhu morfološke procjene, također su napravljeni presjeci za imunohistokemijsko bojenje. Statistička analiza podataka provedena je koristeći MedCalc program za Windows, verzija 19.1.6 (MedCalc Statistical Software bvba, Ostende,Belgija). Za ispitivanje korelacije kalpaina 1 i AR ekspresije korištena je Pearson korelacija. Utvrđena je povezanost određenih kliničko patoloških parametara sa povišenom ekspresijom AR i kalpaina 1, povoljnije preživljenje kod pacijenata sa određenim podtipom tumora, kao i duža stopa preživljenja u razdoblju remisije tijekom pet godina. Takav rezultat može naći svoju primjenu u budućoj svakodnevnoj kliničkoj praksi no isto tako može koristiti kao terapijska meta za određene podtipove tumora. Breast cancer is among the most common cancers in the population of women around the world, affecting a quarter of women newly diagnosed with cancer, and one of the most important causes of death. According to published studies, the incidence of breast cancer varies by race and ethnicity and is more common in developed countries. The aim of this study is to analyze the expression of calpain 1 and AR in breast cancer, to determine the prognostic significance of the expression of tumor markers, to consider whether there is a possibility of using AR and calpain 1 as a potential target response to therapy. The study involved 219 newly diagnosed patients between January 2000. and December 2004. Clinically pathological data were collected using the database of the Department of Pathology. The median age at diagnosis was 65 years. The samples were processed using the microarray method. Tissue blocks embedded in paraffin containing an invasive component of cancer were cut into 4 μm thick sections and stained with HE technique according to a standard protocol for the purpose of morphological evaluation, and sections for immunohistochemical staining were also made. Statistical analysis of the data was performed using MedCalc program for Windows, version 19.1.6 (MedCalc Statistical Software bvba, Ostend, Belgium). The Pearson correlation was used to examine the correlation of calpain 1 and AR expression. The association of certain clinically pathological parameters with increased expression of AR and calpain 1, more favorable survival in patients with a certain tumor subtype, as well as a longer survival rate in the remission period of five years were found. Such a result can find its application in future everyday clinical practice but can also be used as a therapeutic target for certain tumor subtypes. |
