Expression of growth factors and their receptors on human sarcomas. Immunohistochemical detection of platelet-derived growth factor, epidermal growth factor and their receptors

Popis: The expression of both platelet-derived growth factor (PDGF) and epidermal growth factor (EGF), and of their receptors was examined immunohistochemically in a number of normal tissues as well as in benign and malignant mesenchymal tumors. The benign lesions studied were from the skin: palmar fibromatosis (fibroblastic) and from the uterus: leiomyoma (leiomyogenic). The sarcomas included malignant fibrous histiocytoma, leiomyosarcoma, malignant Schwannoma, liposarcoma, fibrosarcoma and synovial sarcoma. In normal tissues the levels of expression of all of the antigens was low, with no apparent coexpression of a growth factor and its cognate receptor. In benign lesions PDGD and EGF were both detectable in combination with their receptor in myofibroblasts. Tumor cells in one out of three leiomyomas were positive for both PDGF and its receptor. Co-expression of PDGF and its receptor was noted in 46% and of the EGF and its receptor in 52% of the sarcomas. Simultaneous expression of PDGF, EGF and their receptors was found in 21% of the cases, mostly malignant fibrous histiocytomas. In some of the tumors the immunostaining pattern was focal and unrelated to a specific cell type. However, in synovial sarcomas, PDGF and receptors for EGF were noted only on the epithelial component. The other two antibodies did not show such a differential staining pattern. The finding in neoplastic lesions of an enhanced frequency of one or two growth factor(s) in combination with their receptors suggests an autocrine model for tumor growth in many cases of sarcomas.
