In August 2015, massive occurrence of Ostreopsis was recorded in Northern Adriatic Sea by scientist from Institute Ruđer Bošković and facebook page is opened for citizen alerts. In parallel, media reported cases of adverse effects in humans on the beaches along the eastern coast. In September 2015, a bloom was recorded in Kaštela Bay where samples for taxonomy and molecular analyses as well as toxins determination were taken. Ostreopsis cf. ovata was identified basing on morphological characterization and ratio of dorsoventral (DV) and anterioposterior (AV) measures. Further, molecular PCR analyses of field samples confirmed the presence of Ostreopsis cf. ovata, with negative results for O. cf. siamensis whereas the concentration of O. cf. ovata in seawater was up to 2.9 x 104 cells L-1 and up to 2.67 x 106 cells g-1 fresh weight of macroalgae mass and 14.4 x 106 cells g-1 dry weight of macroalgae mass. Abundances of C. monotis in seawater were lower than those of Ostreopsis cells (up to 1.1 x 103 cells L-1). In October 2015 O. cf. ovata cells in the seawater decreased up to 1.6 x 103 cells L-1. An ELISA assay and LC-HRMS revealed 6 pg palytoxins per Ostreopsis cell, with a prevalence of ovatoxin-a (3.6 pg cell-1) and lower amounts of ovatoxin-b (1.3 pg cell-1), - d/-e (1.1 pg cell-1), -c (0.2 pg cell-1) and palytoxin (0.1 pg cell-1). |
