Background and Purpose Alyssum montanum L. subsp. pluscanescens (Raim. ex Baumgartner) Trpin (Brassicaceae) is a rare and endangered Croatian stenoendemic plant species growing near Smerovišće only (Samoborsko gorje, southwestern Croatia). The aim of the study was to investigate A. montanum subsp. pluscanescens micropropagation possibility in order to contribute to biodiversity protection programme. Material and Methods Shoots originated from aseptically germinated seeds collected on natural habitat were used for culture initiation. Multiplication rate of shoots was invesigated during 4 subcultures on half or full strength MS medium containing 2.9 ; M GA3 and different concentrations of BA or without BA. Multiplicated shoots were elongated on MS medium with 0.2 ; M BA and 14.5 ; M GA3 during 4 weeks. Elongated shoots (5-7 cm) were cut off and rooted on full strength MS media containing three different kinds of auxins in various concentrations. Rooting was monitored during 2 subcultures. Results and Conclusions The sterilization of seeds was successful. After 4 weeks all cultures remained sterile and all the seeds germinated. The higest multiplication rate per explant (2.6 in first and 2.8 in second subculture) was achieved on full strength MS medium supplemented with higher concentrations of BA (0.5 and 1.0 ; M, respectively). The best rooting was observed on media supplemented with 2.5 and 4.9 ; M IBA. These results indicate that Alyssum montanum subsp. pluscanescens could be succesfully preserved through micropropagation. |
