Abstrakt: This paper describes the equipment and procedures that are necessary for the successful reconstitution of an ion channel into a planar bilayer. Included are techniques for the preparation of the protein, the formation of the lipid bilayer, the fusion of vesicles with the bilayer, and data acquisition and analysis. As an example, the reconstitution of the ryanodine receptor/Ca2+ release channel using the planar bilayer technique of spreading from bulk lipid solutions is discussed.Copyright 1994, 1999 Academic Press
