Purification of Rat Retinal Ganglion Cells by Immunopanning

Abstrakt: A simple two-step "panning" procedure that allows the isolation of up to 50% of the retinal ganglion cells from postnatal rat retina to greater than 99.5% purity is described. Rat retinal suspensions are prepared enzymatically and passed sequentially over two antibody-coated petri dishes. The first dish is coated with an antibody that selectively depletes the retinal suspension of macrophages and endothelial cells. The second dish is coated with a Thy-1.1 antibody which selects the retinal ganglion cells. The purified retinal ganglion cells are recovered from this dish enzymatically and can then be used for further biochemical, molecular biological, electrophysiological, or tissue culture studies.
