In this study, Crain et al. describe the impact of histone H4 lysine 20 monomethylation (H4K20me1) on gene expression in Drosophila, showing that Set8, the histone lysine methyltransferase responsible for H4K20me1 deposition, and H4K20me1 reader l(3)mbt regulate gene expression and occupy chromatin through H4K20me1-independent mechanisms. These findings indicate that H4K20me1 does not have a significant role in regulating gene expression and may be dispensable for the function of Set8 and L(3)mbt in development and disease. |
