The neurotoxicity of misonidazole: potential modifying role of phenytoin sodium and dexamethasone

Abstrakt: The dose-limiting effect of the hypoxic-cell sensitizer misonidazole is neurotoxicity, manifested primarily by peripheral neuropathy and in some patients by ototoxicity and encephalopathy (Dische et al., 1978; Wasserman et al., 1980). There has been general agreement throughout the world that the total dose of misonidazole, expressed as g/m2, is the most important factor in the incidence of neuropathy. Other factors affecting the incidence of neuropathy may include the time over which the drug is given and the hydration of the patient, but these have not been clearly established (Dische et al., 1978).
