Symptoms of mosaic, with puckering and blistering on leaves, were found in cultivated melon in 2020 in Bali, Indonesia. RT-PCR, a universal primer of CI gene of Potyvirus, successfully amplified a DNA sized ± 683 bp. The homologies of nucleotide (nt) and amino acid (aa) sequences of PRSV-W Bali isolate to those isolates from other countries, was 94.1–98.8% nt and 94.6–99.1% aa. The cylindrical inclusion nucleotide and amino acid sequences of three isolates, from Bali, had the highest identity with that of PRSV-W isolate cucurbit in East Timor. PRSV-W Bali was in the same cluster with PRSV-W from East Timor and Australia. By mechanical inoculation and aphid transmission, the virus was able to infect systemically melon plants. This is the first report of PRSV-W infection on melon in Indonesia. |
