It was found that 8-10 % of 3H-glutamic acid (GLU) taken up into striatal slices were converted to 3H-GABA (newly synthesized H-GABA) during the incubation in the presence of nipecotic acid (5×105M). The high potassium (60 mM)-evoked and Ca2dependent release of newly synthesized 3H-GABA was found to be more prominent than that of accumulated 3H-GABA from the incubation medium. The evoked release of 3H-GABA from striatal slices was significantly suppressed by in vitroaddition (10−5M) of δ-aminolevulinic acid (DALA) as well as some GABA agonists such as imidazoleacetic acid and muscimol (MUS). |
