Thiolysis of Dinitrophenylated Myosin with Restoration of Adenosine Triphosphatase Activity

Abstrakt: When rabbit skeletal myosin, which had incorporated 1.43 moles of 1-fluoro-2,4-dinitrobenzene per 5 x105g of protein, was treated with 0.05 m2-mercaptoethanol at pH 8.5, complete restoration of the K+(EDTA)- and Ca2+-ATPase activities occurred. Concomitantly, 0.73 mole of the dinitrophenyl group was removed. This was established from incorporation data and difference spectra obtained for dinitrophenylated myosin treated with and without 2-mercaptoethanol.
