The objectives of the present study were to (1) diagnose cytological endometritis (CYTO) during a timed AI protocol and assess the presence of endometrial polymorphonuclear cells (PMN); (2) to determine the relationship between PMN and size and position of the reproductive tract in postpartum multiparous Nellore cows and its effect on pregnancy outcomes. A total of 201 cows were enrolled. A standard estrus synchronization protocol (TAI, d 0) was used. Endometrial cytology samples were taken (days postpartum 28 ± 61) by cytobrush, and size and position score (SPS) of the reproductive tract was assigned on the first day of the protocol, d -11. For each slide, 300 neutrophils and epithelial cells were counted in 3 different locations by 2 operators and the polymorphonuclear cell ratio (% PMN) was assessed (CYTO-positive ≥ 5% PMN, CYTO-negative < 5% PMN). Reproductive tract scores were assigned as SPS1 as small and compact resting within the pelvic cavity; SPS2 as intermediate, resting partially outside the pelvic cavity; and SPS3 as larger and resting mostly outside the pelvic cavity. Pregnancy diagnosis was performed by transrectal ultrasonography 30 days after TAI. Data were analyzed with PROC GLIMMIX with a binary distribution and a logit link; and PROC FREQ model in SAS 9.4. Overall pregnancy rate was 44.3 % (n= 89/201). Cows presenting PMN higher or lower than 5.0 % had no statistical difference (P=0.2) when comparing pregnancy rate. However, the probability of a CYTO-positive cow becoming pregnant (29.4 %; n= 5/17) was 16 % lower when compared to a CYTO-negative cow (45.6 %; n =84/184; P<0.05). There was no significant relationship between SPS and CYTO positives and negatives cows (P=0.67). In conclusion, about 8.5% of inseminated cows were positive for uterine disease which led to a lower probability of these cows becoming pregnant. |
