The goal of this study was investigation of the effect of glutathione in a soybean lecithin-based semen extender on post-thawed buck sperm quality. Semen samples (n=20) were collected from four Mahabadi Bucks (five replicates) and diluted with four extenders after pooling: 1) egg yolk tris-based extender (EYT), 2) soybean lecithin-based extender (L0), 3) soybean lecithin-based extender containing 5 mM glutathione (L5), and 4) soybean lecithin-based extender containing 10 mM glutathione (L10). The sperm motility and moving characteristics, percentage normal sperm, plasma membrane intact sperm, sperm viability, and the levels of malondialdehyde were determined after freezing-thawing process. Glutathione at the level of 10 mM provided the lowest percentage of total and progressive motility. Percentage of plasma membrane intact sperm was lower in L5 and L10 groups than EYT and L0 groups. Sperm viability was significantly decreased in L10 group in comparison to EYT group. Also, percentage of normal sperm was not significantly different between groups. The results of this study show that the most post-thawed sperm quality characteristics were not significantly different between EYT and L0 groups. Also, addition of 5 and 10 mM glutathione in a soybean lecithin-based extender can not improve buck sperm quality after freezing-thawing process. In conclusion, it seems that soybean lecithin-based extender can be a suitable replacement for egg yolk tris-based extender, and addition of 5 and 10 mM glutathione can not fortify the soybean lecithin-based extender for buck sperm cryopreservation.
