SEPARATION OF HL-A TRANSPLANTATION ANTIGEN SPECIFICITIES

Abstrakt: Soluble HL-A transplantation antigens, isolated from spleens of tissue-typed donors, have been examined by diethylaminoethyl ion-exchange chromatography and the several specificities expressed by each individual were shown to be present on separable molecules by examining the peak positions of each specificity in the column fractions. This was done by inhibition of platelet complement fixation or of cytotoxic reactions using well defined HL-A typing sera. A tentative beginning has been made to map the sequence of elution of the molecules carrying different HL-A specific determinants. There is evidence against the view that the products of the first HL-A sublocus have any family relationship as seen by diethylaminoethyl adsorptive capacity. For the second sublocus, there are insufficient data to draw such a conclusion. Certain non-HL-A (leuco) platelet antigens eluted from the columns quite separately from those determined by the HL-Alocus. HL-A antigens having the same properties as those isolated from human spleens can also be obtained from lymphoid cell suspended tissue cultures established from peripheral leucocytes of tissue-typed donors.
