An indoloquinoline compound prepared from lysergic acid base has been found to have an α-blocking action on in vivoand in vitropreparations. The compound reverses the blood pressure response to epinephrine from pressor to depressor in pithed and decerebrate cats. In renal hypertensive dogs, the compound produced a significant sustained decrease in blood pressure. on rat vas deferens preparations, the spasmogenic effect of epinephrine and norepinephrine was blocked by the compound similar to, but not in the same degree as that observed with dibenamine. The primary effect of the compound appears to be an interference with sympathetic transmission postsynaptically at the α-receptor sites resulting in peripheral dilatation. |
