A rapid and sensitive ultra performance liquid chromatography (UPLC) method was developed to quantify the principle active components (o-chlorobenzamalononitrile, nonivamide, dihydrocapsaicin) in a variety of aerosol defense sparys obtained from independent distributors of the products. Chromatographic separation of the target ingredients was achieved using UPLC employing a reversed phase C18 column and a stepwise gradient of methanol and distilled water. Quantification of the analytes was achieved using ultraviolet detection at a wavelength of 229nm.The plot of concentration versus peak area was linear over the range of 1.0-100.0ug/ml with a correlation coefficient (r2)≥0.999 for each analyte. The accuracy of the assay ranged from 97% to 105% for all analytes and the quality control samples at 10, 50, 100ug/ml. The inter-assay and intra-assay precision of the method were achieved with a relative standard deviations lower than 3.6%. The results of the analysis demonstrate the degree of variability in the active ingredient content of the sprays. |
