In order to avoid time-consuming desalting by ion-exchange column-chromatography a two-layer plate is proposed for TLC of urinary amino-acids. A small stripe of the plate contains 10% of a strongly acidic ion-exchanger in cellulose, the remainder is covered with pure cellulose. Urine is applied onto the exchanger layer. Neutral and acidic substances are washed from the layer by water. The amino acids are developed into the pure cellulose layer by methanol/ammonia/water (50:15:35). After cutting off the exchanger stripe, the amino-acids are developed twice by n-butanol/acetone/ acetic acid/water (35:35:10:20). As demonstrated by examples, the method is suitable for screening tests for diseases concerning the urinary amino-acids. |
