Electron microscopic visualization of membrane Ca2+-binding sites in barley root tip cells: Inductionin vivo by seed germination in the presence of CaCl2

Abstrakt: Summary Germination of barley seeds in the presence of 1 mM, 10 mM or 50 mM CaCl2 induces formation of electron-dense membrane-associated globules, whose structure and distribution closely resemble those observed using the cytochemical Oschman-Wall's technique. Treatment of thin sections by pronase and phospholipase A solutions lead to a partial dissolution of intramembranous dense material. The images obtained are similar to those of samples, germinated in distilled water but subjected to calcium-supplemented fixation and washing procedures during tissue processing for electron microscopy.
