Effect of product inhibition on kinetics of drug elimination

Abstrakt: In view of recent indications that drug biotransformation processes may be inhibited in vivoby the products of such processes, pharmacokinetic studies have been carried out to determine the possible effects of product inhibition on drug elimination. Theoretical considerations and digital computer simulations show that an apparent increase in the biological half-life of a drug with increasing dose may result from product inhibition if the dissociation constant for the drug metabolite-enzyme complex is appreciably lower than the Michaelis constant for the drugenzyme complex, if drug metabolite levels remain relatively constant for some time due to slow elimination of the metabolite, and if the level of drug in the body does not appreciably exceed the apparent in vivoMichaelis constant.
